Cell proliferation markers in the transplanted canine transmissible venereal tumor Arq. Bras. Med. Vet. Zootec.
Santos,F.G.A.; Moro,L.; Cassali,G.D.; Paixão,T.A.; Campos,P.P.; Silva,S.S.; Vasconcelos,A.C..
Adult male mongrel dogs were subcutaneously transplanted with the canine transmissible venereal tumor (TVT) on the hypogastric region. Twelve specimens of tumors were collected, half during the proliferative phase and the other half during the regressive phase. Fragments of the tumor were fixed in 10% buffered formalin and routinely processed for light microscopy. Sections of 4µm were stained by Schorr or AgNOR or either immunostained for MIB1 (Ki67). Schorr stain, AgNOR and MIB1 showed an increased proliferative activity through mitotic index, nuclear argyrophilic protein stain and cycling tumoral cells in the growing tumors, respectively. All of the three cell proliferation markers were able to distinguish the TVT in both evolution phases. MIB1...

Comparative study of DNA synthesis and nucleolar organizer regions of sinusoid littoral cells in mouse regenerating liver Biocell
Martín,Carlos A.; Surur,José Miguel; García,Marcela N.; Corrons,Félix; Badrán,Amado F..
Variations in DNA synthesis (DNAs) and Nucleolar Organizer Regions (NORs) were studied in the littoral cell population from regenerating liver of C3HS inbred mice standardized for periodicity analysis. Immunohistochemical detection of Bromodeoxyuridine (BrdU) with a monoclonal antibody and silver staining of NORs (AgNORs) were assessed by means of a digital image analysis system in histological sections. Tissue samples were obtained every four hours from the 30th to the 54th hours after a partial hepatectomy. The results showed, in both parameters, a gradual increment of the values during the period studied, with highest values (DNAs 107.1 ± 16.1 SE; AgNORs 77.3 ± 3.4 SE) located at 16:00/54 Time of Day / Hours Post- Hepatectomy (TD/HPH), which were...

Cytogenetic characterization of a population of Bryconamericus aff. iheringii (Characidae,Tetragonopterinae) Genet. Mol. Biol.
Paintner-Marques,Tânia Regina; Giuliano-Caetano,Lucia; Dias,Ana Lúcia.
A cytogenetic analysis of thirteen specimens of Bryconamericus aff. Iheringii revealed a diploid number of 52 chromosomes and a karyotype of 8M+22SM+10ST+12A with a fundamental number (FN) of 92. The nucleolus organizer regions (NORs) were studied by means of AgNO3 and CMA3 staining as well as by FISH using an 18S rDNA probe. NORs were found to be located at the terminal position on the short arm of the submetacentric chromosome pair. C-banding showed strong telomeric and centromeric staining in the majority of the chromosomes, and a similar pattern was observed after treatment with AluI restriction enzyme

Diversity and chromosomal evolution in the genusAncistrusKner, 1854 (Loricariidae: Ancistrini) from three hydrographic basins of Mato Grosso State, Brazil Neotropical Ichthyology
Mariotto,Sandra; Centofante,Liano; Moreira-Filho,Orlando.
Cytogenetic analyses were carried out in 117 specimens of seven species of the genus Ancistrus from three hydrographic in Mato Grosso State: Paraguay, Araguaia-Tocantins and Amazon basins. Conventional cytogenetic techniques were used to obtain mitotic chromosomes. C-banding was performed to detect heterochromatic regions and silver nitrate staining was used to identify nucleolar organizer regions (Ag-NORs). The counted and paired chromosomes revealed diploid numbers ranging from 2n = 40 to 2n = 54 with karyotype formulae varying from FN = 80 to FN = 86. Single marks in distinct chromosomes identified the nucleolar organizer regions. The constitutive heterochromatin was scarce in the diploid number from 2n = 50 to 2n = 54 and conspicuous blocks were...

GOSSLER,VANESSA S.A.; SANTOS,FRANCISLAINE A.G.; AZEVEDO,ALINE R. DE; GONÇALVES,PRISCILA C.; RIGOLO,HUGO A.; TREVISAN,CLEONICE; MASSENO,ANA PAULA B.; CARDOSO,ANA PAULA M.M.; PAPA,PAULA C.; CASTILHO,CALIÊ; GIOMETTI,INES C..
ABSTRACT This study aimed to determine the histological features of the endometrium of bitches, as well as the cell proliferation at specific moments of diestrus, 10, 20, 30, 40, 50 and 60 days post ovulation, correlating the endometrial thickness with the uterine cell proliferation and the metabolic state (weight, blood glucose and plasma cholesterol) of the animals. Therefore, the right and left uterine horns of 26 clinically healthy bitches submitted to ovariohysterectomy were histologically analyzed 10, 20, 30, 40, 50 and 60 days post ovulation. The hematoxylin-eosin and AgNOR staining techniques were performed. All parameters were evaluated by ANOVA and post-hoc Tukey test (p<0.05). The correlation between endometrial thickness and uterine cell...

Interindividual size heteromorphism of NOR and chromosomal location of 5S rRNA genes in Iheringichthys labrosus BABT
Carvalho,Rafael Augusto de; Dias,Ana Lúcia.
Twenty-five specimens of Iheringichthys labrosus from the Capivara Reservoir were analysed cytogenetically. AgNORs were detected in a pair of ST chromosomes, in the telomeric region of the long arm. Some individuals showed size heteromorphism of this region between homologous chromosomes. Treatment with CMA3 displayed GC-rich regions corresponding to the AgNOR pair, plus other fluorescent staining. In situ hybridization by fluorescence (FISH) with the 18S rDNA probe showed only one pair of stained chromosomes, confirming the heteromorphism observed with AgNO3 and CMA3 in some individuals. The 5S rDNA probe revealed telomeric staining on the long arm of a pair of chromosomes of the ST-A group, probably different from the NOR pair.

Marcadores de proliferação celular na avaliação do crescimento do tumor sólido e ascítico de Ehrlich Arq. Bras. Med. Vet. Zootec.
Silva,A.E.; Santos,F.G.A.; Cassali,G.D..
Twenty BALB/c mice were inoculated with cell suspension of Ehrlich tumor. Ten mice were inoculated in the cushion plant (solid form) and the other 10 in the peritoneum (ascitic form). Animals were euthanized on different times (7 and 14 days). Cytological and histological slides, immunohistochemical (PCNA) analysis and NORs silver impregnation technique were performed. The results showed more proliferation on the 7th day in the ascitic form and on 14th day in the solid form, using both analyses (PCNA and AgNORs). The alterations observed in the Ehrlich tumor’s proliferation activity suggested that the growth curve is different between ascitic and solid forms. In the first one, the proliferation peak occurs on the seventh day and in the solid tumor the...
